Study of optimal conditions for invertase production from a local isolate of Candida spp

Abstract

A preliminary screening process was conducted on a number of yeasts isolated from different sources in terms of their ability to produce the enzyme invertase. It was found that five isolates of them belonging to the genus Candida were distinguished by their ability to produce the enzyme. The isolate coded C5 was the most efficient of those isolates. The optimum conditions for enzyme production were studied using the submerged culture method of this isolate. It was found that the highest productivity was achieved by using raffinose as a carbon source, at a concentration of 3%, and a mixture of yeast extract and ammonium sulfate, at a ratio of 1:1, at a concentration of 0.4%, as a nitrogen source, with an initial pH of 6, and an incubation period of 28 hours at 35 °C. The enzyme activity under these conditions was 1.58.
unit / ml, with a specific potency of 6.07 units / mg.
